I feel not competent enough to answer your question, but I would like to give you one consideration in pursuing your described purpose.

There are some thoughts about looking at (phospho)lipids in a simplified way.

The rationale behind a particular structure (bilayer, micelle or non-bilayer) is explained by the so-called shape-structure concept of lipid polymorphism (P.R. Cullis, B. de Kruijff, Lipid polymorphism and the functional roles of lipids in biological membranes, Biochim. Biophys. Acta 559 (1979) 399– 420).

In essence, the message is:

Lipids have an overall cylindrical shape when the cross-sectional area of the lipid headgroup is similar to the cross-sectional area of the acyl chains and they’ll tend to form bilayers. This is for example the case for phosphatidylcholine (PC).

If the cross-sectional area of the headgroup is larger than that of the acyl chains aggregate structures such as, lyso-phospholipids, with a positive curvature are favoured and they’ll tent to form micelles.

If the cross-sectional area of the headgroup is smaller aggregate structures with a negative curvature are preferred, so-called hexagonal phase (HII) or cubic phase.

In other words: My remark comes down to the warning that you want to create a bilayer of a type of lipid that has a tendency to form a non-bilayer membrane.

Anyway, hope this is of any use to your work.

Rob is right. Lysophospholipids contain only one fatty acid chain rather than two. They act as detergents and tend to form micelles, not double membranes. If you add lysolipids to membranes, solubilisation occurs (see doi:10.1590/S0100-879X2002000700001 for a review on solubilisation).

Engelbert Buxbaum I think the preferred way is to assume the lysophospholipid as a ligand and then add (required number) to the bilayer membrane of phospholipids. This way we do not alter the characteristics of the membrane and still we have what we want in the membrane.

Even traces of detergent alter the fluidity of membranes and hence the activity of transmembrane enzymes. Usually, you see an activation at very low concentration, followed by an inhibition as you approach solubilising concentrations. If you increase the detergent concentration further, you may even get inactivation due to removal of anular lipids. Rigaud's review discusses this in detail.
