I am interested in using mass spec to quantify steroid hormones in human saliva samples, but I am completely new to mass spec. Unfortunately, there seem to be very few protocols for this, and given that steroid hormones are found in very low quantities in saliva, it makes their quantification that much more difficult.

Does anyone any advice on the following:

- How to prepare frozen saliva samples an absolute quantification of steroid hormones?

- Are there any matrices that can be used to mimic saliva for use in the standard curve?

- Does anyone have a protocol for doing anything similar to this?
