i had conducted quantitative assay of phosphate solubilization and estimated the solubilized phosphate with change in ph at 3rd day 5th day 7th day and 9th day for 11 bacterial isolates. now i have to graphically represent the obtained data using two y-axis line graph( concentration of solubilized P on y-axis, days on x-axis and pH on another y-axis).
suggeste me any application and some steps so i can create graph.
You can do this in Excel within the Microsoft Office applications, if you have it. You just need to make two columns of numbers for the x and y values, select them, and choose the style of "chart" you want.
Hello Manisha, I see you have three independent variables (IVs) and only one response variable; a typical case of a univariate analysis. You have 11 isolates which i think should be reduced to the most relevant ones (say 4 or 5), different pH levels (number unknown) and time (days) 3, 5, 7 and 9. It means phosphate solubilization depends on the nature of isolate, pH and duration of incubation. You need to plot one dependent variable (phosphate solubilization) against the 3 independent variables in a single graph. Well then, you have to create a single plot with 3 y-axes each for an independent variable. Researchers generally prefer to have dependent variables plotted on y-axis and independent variables on x-axis, however, in a multilinear relationship like this, i think you could be given the benefit of the doubt. Because of the problem of scaling, i doubt if one single plot will do. A univariate analysis in SPSS returns 3 separate plots for each independent variable. However, you could attempt one single plot in Matlab and then add the axis manually. Personally i would go for a clustered bar graph after reduction in the number of isolates.

That graph is a bit more complicated because it has two different y-axes. In Excel, you have to find the command to add another axis to an existing graph with just one. Your data can conveniently be organized as pairs of columns, with each pair containing the x and y values. You will then select the columns of numbers for x and the particular y axis to add each line.
Many people use GraphPad Prism or SigmaPlot for preparing graphs.
You can use Matlab and upload an excel workbook containing your data in columns. The advantage of Matlab as opposed to excel is its ability to display and manhlpilste data with regression tools.
