Blood from some donors are fatty, after centrifugation an upper layer forms which is difficult to remove, in order to get clean plasma for cfDNA analysis.

maybe you could try piercable centrifugation tubes and simply remove your plasma-aliquot with a Syringe from beneath the fat-layer.
