Why does my sample shine too bright green all over the whole cell? It's overexposure of FITC I believe. The green colour should shine bright green at the cell lining (oocyst) only as what obtained by CDC.
I have followed the method by FITC manufacturer. The staining method should not be the problem. Can anyone comment?
Check your image acquisition software. There is a panel where you can adjust the exposure settings during imaging. You can reduce the time manually, or set the settings to Automatic, so that the image is not overexposed. If you are quantifying your images, take note of this exposure time, so that the next time you do imaging, you will use the same exposure time.
