I am performing an experiment where MDM are infected at a certain multiplicity of infection (MOI) 4 days after GM-CSF differentiation. I would like to be accurate in the MOI I am using to infect.
Do you know a method to measure the quantity of MDM in the plate without cell detachment? Or do you know whats the percentage of viability of macrophages since they are seeded in the plate and treated with GM-CSF for 4 days?

CytoSMART makes live cell imagers that take images and use image analysis to quantify cells while they are in the plate, in the incubator. it could track your experiments over time so you can see the changes in cell number and morphology over he 4 days.

I dont have an answer to your question but I have always had a great interest in the role of macrophages in the processing of antigens. Have written a theoretical on the subject that can be found on Researchgate.
Macrophages were first discovered to elaborate immune RNA that could elicit antibody formation in B lymphocytes. May I ask what you are searching for with your experiments?

You could quantify DNA content in cell extracts at the end of the experiment so that you can normalize for cell counts across the groups you are evaluating.
