Hello, fellows, I would like to measure soil enzymes activity such as urease, dehydrogenase and alkaline phosphatase. I already have all the necessary references for the protocol. However, I would like to know whether I will need to prepare standards and if someone could help me with a link to prepare enzymes standards. Or do I only need to only extract enzymes and determine optical density on absorbance spectrometer? For instance, when determining soil available P, I often prepare standards of 0, 0.4, 0.8, 1.2, 2.0 ppm, is that also application in the determination of soil enzyme activity. Your advice is highly appreciated. Thank you.
