I am running an experiment on the degradation/removal of oxytetracycline and for the analysis, I am using UV-detector HPLC system with C18 column, 355nm wavelength. My mobile phase is 60/25/15 v/v of oxalic acid, methanol, and acetonitrile. For the methanol and acetonitrile, I suppose that I just need to measure it into desired volume, but for the oxalic acid, I do not know how to prepare it into liquid solution as my lab only have the solid only. Your help is much appreciated.
You must prepare a certain concentration of oxalic acid in distilled water similar to the buffers used in HPLC. In other words, the third component of your mobile phase is distilled water containing oxalic acid.
Thank you so much for the answer. I have found the concentration of the oxalic acid, but I am not sure if it also applies to the methanol and acetonitrile. The article that I am referring to wrote "The mobile phase used as eluent was 0.01mol/L oxalic acid:methanol:acetonitrile (67:11:22)." Does the methanol and acetonitrile also diluted to 0.01mol/L in this context?
You can use multiple containers to enter HPLC. Insert one component into each container and then specify the ratio in the HPLC software according to the container number.In one of the container, pour the oxalic acid solution at a concentration of 0.01 M.
