I am working with cardiomyocytes (CMs) that differentiate from pluripotent stem cells. I am currently struggling to obtain single cardiomyocytes for flow cytometry analysis. I used 10X TrypLE for 10 minutes, and although it made the CMs single, we observed a high number of dead cells in the flow cytometry analysis. I also tried 5mM EDTA; however, its effectiveness was variable. Since I keep CMs long-term, it is particularly challenging to make them single.

Does anyone have any tips or a user-friendly protocol to achieve single CMs with high viability for flow cytometry?
