It depends upon the nature of the sample you have. If you have liquid sample it's good to disperse one drop of sample in Nickel or other grid. HRTEM requires much more clean sample to observe lattice fringes clearly.
For HRTEM study,your sample should be very clean.The polysachharides may interrupt for lattice fringe study.If you have used sugar for nanoparticles synthesis only and have no further application of polysachharide,you should removed and washed it before HRTEM through sonication and centrifugation.Nobody can say how much dilution you should take without knowing the concentration.first wash your sample with ddH2Ol,make suspension and load one drop of sample on the grid.
