Suspend 0.5 to 1.0% (w/v) of mucin in Agar based medium. During autoclaving the medium shall become quite homogeneous. Use it for for direct isolation of mucin degrading microbes. Prior to this approach you may need to enrich culure using mucin containg broth.

The dissolved mucin was dialyzed overnight against three changes of 5 L phosphate buffer using dialysis membrane with a 2300 molecular weight cut off (Spectra/Por 7 Dialysis Membrane; Spectrapor). Mucin was added to media to a final concentration of 0.5% and filter sterilized (Stericup 45μm).

Hi Krishan Selwal, I am trying to make a medium containing mucin as well, but i noticed that after autoclaving the mucin there are still bacteria present (visable with a microscope). They do however not grow on TSA or in LB. Did you encounter this problem? I allready tried the Type II and III porcin gastric mucin and both seem to have the same contaminant. I am concerned that by filtration the mucin would get stuck in the membrane of the filter, so i didn't try this yet.

Well, I didn't encountered with such problem. It is quite possible that you might not have purified mucin. I normally purified type 2 mucin before adding in BHI and MSA medium.
