Hi everyone, I’m planning to prepare an L-DOPA solution for cell culture. According to the data sheet from Sigma, the solution should be freshly made because L-DOPA oxidizes quickly when exposed to air and light, turning dark.
I’m wondering, is it necessary to add ascorbic acid or fill some inert gas (eg: N2) to prevent this oxidation? If yes, do you have any protocol?
Any practical tips on preparing and handling L-DOPA solutions during experiments would be greatly appreciated!
Adding ascorbic acid as a reducing agent can inhibit the oxidation of L-DOPA, prolong the stability of the solution, and reduce color changes and loss of activity. Protocol
3. Sterile filter the solution through a 0.22μm filter to remove any particulate matter.
For instance, if I must prepare 10μM solution, I would need 197.19 x 10 x 10^-6 g/L = 0.00197 g/L (approximately 0.002mg/ml or 2μg/ml),
(Molecular weight of L-DOPA is 197.19 g/mol).
I would dissolve 0.02mg of L-DOPA in 10ml of culture media. I would add ascorbic acid (0.1%, w/v) and filter sterilize. I would protect the solution from light.
Some points to be noted:
1. Prepare L-DOPA solution fresh on the day of use to minimize degradation. Do not store aqueous solutions of L-DOPA for more than one day.
2. If you need to store it for a short time, store the prepared solution in a dark bottle or container, or wrap the container in aluminium foil to protect it from light at 4°C.
3. Monitor the solution for any signs of darkening or precipitation, which could indicate oxidation. If you observe these changes, discard the solution and prepare a fresh one.
