Hi everybody,

I want to investigate the bacteria-mucus interaction by using an ex vivo model or an in vitro model.

To prepare intestinal mucus is the very crucial step.

I have checked several references, finding many methods (as following), but still, have no idea which method is optimal to mimic the mucus layer that attached on the intestinal epithelium.

Related mucus preparation or simulation method and my concern are listed:

(1)Scrapping methods.

The mucus mechanical structure could be destructed, especially the inner layer of mucus.

(2)Mucus secreted by cell lines (HT29, caco-2, etc.,.).

The inadequate secretion

(3)Commercially available mucins (bovine submaxillary mucin or Mucin from porcine stomach).

The mucins from submaxillary or stomach are different from the ones from the intestine.
