I would like to perform an MTS assay using the following concentrations: 100uM, 50uM, 20uM, 10uM, 5uM, 2uM, 1uM and 0.5uM. I will be adding 100ul of each of my drug doses to a well on a 96 well plate already containing my suspension cells in 100ul of medium. The final volume of my drug dilutions will be 4ml.
I have purchased 5mg of my drug, looking at the data sheet the highest concentration of stock I can make is 10mM by adding 0.9735ml of DMF and dissolving.
Can anyone please help with the calculations for how to make my final concentrations from my stock? (Maths is not my strong point and I am getting easily confused)

Once you make 10mM (=10000 uM) solution you should the following dilutions:
1-for preparing100uM solution:take 1 part of the stock solution (10mM) and dilute with your solvent upto 100.
for preparing 50uM solution: take 0.5 part of the stock solution (10mM) and dilute with your solvent upto 100.
For preparing 20uM solution: take 0.2 part of the stock solution (10mM) and dilute with your solvent upto 100.
and so on .......
