I have some polyvinyl alcohol hydrogel samples with silver nanoparticles, which I need to analyze with TEM in order to see the size/morphology of the nanoparticles. Since I do not have an option to use microtome, I decided to dissolve the samples in water (I used 1:5 ratio by weight), drop them on a Cu grid and evaporate the solution, hopefully that way the nanoparticles will remain on the grid to be viewed on TEM. However, I’m having problems with this method, as after 24 h drying, probably a thin polymer film formed on the grid, apparently too thick and completely non-transparent for the TEM beam, and there is no visibility at all. If anyone has experience with similar samples, I would greatly appreciate any advice on how to prepare the samples (e.g. what dissolution ratio, how much solution to add on the grid etc.) for transmission electron microscopy.
