We dried leaves of Curry plant for 10 days under shade, made powder in blender and incubated for 48 hours. After incubation solution was filtered with whatman filter paper.....Filtrate was used for phytochemical analysis but all test got negative for flavonoids, alkaloids, etc. How can we get proper results ??
You cannot expect much flavonoids in the hexane extract. Please use a relatively polar solvent for extraction. Eg. 80% methanol containing 0.1% formic acid or acetic acid.
as is hexane can only extract fat/wax only you need to develop proper solvent system
Dear Santosh, please be clear with your objective of research... so that clear answer will be expected., as fatty matter can be extracted by hexane.... there cannot expect flavanoids and/or other phyto-compounds except some terpenoids like volatile oils, waxy and oils etc. If you want to extract all phyto-chemicals from the curry leaves both ways you can first you may proceed with High polar solvent such Methanol or Ethanol and another way you can extract with increasing polarity n-hexane, Chloroform, Ethyl acetate and ethanol is the systemic way to isolate and /or to extract the components on polar based.

Thank you all, but I tried ethanol , water and chloroform for Prosopis juliflora and got negative results for terpenoids and quinones which I had observed positive in articles
