We are currently inducing osteogenesis, and for the osteogenic medium we need the concentration of β-glycerophosphate to be 20 mM in the final volume of the media (Vf = 50 mL). Therefore, we tried to prepare a 20 M mother solution, but we faced 2 problems:
1. The mass of β-glycerophosphate is so huge compared to the volume of the solvent
2. 100% EtOH did not dissolve β-glycerophosphate
Please find below our calculations:
β-glycerophosphate = 20 mM (216.04 g/moL)
- Mother solution = 20 M and Vf = 50 mL => V1 = 50 µL
- Prepare the soltuion in EtOH and V = 200 µL => n = 0.004 mol => m = 864.16 mg
Why do you need the stock solution to be 20M? You are probably facing a solubility issue at that high concentration. If the stock solution was 500mM (in water) and you wanted a final concentration to be 20mM, you would need to add 2mls of BGP. 2mls into 50mls is not going to alter the concentrations of any other factors or media components significantly enough to affect osteogenesis.
Dear Tom Masi thank you for your answer. I will prepare 500 mM stock solution, since you confirmed that the addition of 2 mL into 50 mL does not alter the concentration of other components of the media.
