I want to perform Wright’s stain on keratinocytes growing in plastic 24-well plates, but I am not sure if I can fix the cells air-dry completely, or I should do some kind of chemical fixation? Would it work in plastic plates?
Remove the media, wash with PBS and air dry it before staining with Wright stain! if you are worried about complete air drying, use methanol to fix the cells and proceed with Wright staining!
Note: Wright stain is made with methanol, so there is no need to fix the cells, But if you are worried about water artifacts it is better to fix with methanol!
