Is there any method through which we can perform MTT assay for different time points in a single 96 well plate. I'm treating cells with six different concentration and now want to see there cytotoxic effect at 3, 6, 12 and 24 hours in a single 96 well plate as we are short of these plates and my PI have no money to buy them, so, I will have to utilize them carefully.
Dear Sandeep, all your problems will be solved if instead of MTT you use Alamar Blue. With this similar assay you can run kinetic at different time points and the results (at least for me) were very similar to the one I got using MTT. it is very easy;
you can cover the cells with aluminium foil and test them in certain time.
my suggestion is if you use one plate for one selected time will be best. generally plate have to put during reading without lid .
