I am planning to run an experiment which comprises IHC or IF staining of melanocytes (MART-1 or SOX-10). FFPE samples of the skin are the target tissue. Subsequently I will perform laser capture microdissection (LCM, PALM Zeiss) and miRNA expression profile analysis in the dissected compartments.

Literature mining revealed IHC or IF staining the most degradative part of whole procedure. Antigen retrieval seems to be the key step.

Do you have optimized protocols/preferred kits for the IHC or IF staining which limits RNA degradation, especially when conducting LCM?
