I have been trying to patch leukaemic cells which are non adherent . Tried different coating agents, but no success.Confused with what to be tried to make the floating cells attach.
I would suggest that you use multiple coating. Say if you are using poly-l-lysine, what you do is apply the poly-l-lysine solution on to the surface of a very-clean coverslip of your choice. Let it dry for a few minutes and then re-apply poly-l-lysine again, let it dry and then re-apply for the third time. Once it is dried properly then you can place a drop of solution contain your cells on the coverslip. Let the cells settle down in a vibration-free (or least in a calm) environment at least overnight. Make sure that the solution does not evaporate and the cells do not dry out.
