Chemical vapor deposition (CVD) was used to grow graphene on copper (Cu ) substrate, then the single layer graphene was transfered onto a PDMS substrate. I want to measure the bonding strength of the transfered graphene on PDMS substrate, what should I do? Thanks.

I was thinking about this and initially was thinking that the question was wrong. That maybe this was to be calculated and not measured. Well, I think this can still be calculated but I realized this should be measured okay with AFM. Then I went and found the below article that uses AFM as a "nano-scratch technique" where the researches apply a lateral force at the interface of the graphene and the substrate in order to debond the graphene layer from the given substrate surface. There should be many more articles like this, but I think this could be a start for you to try using AFM if your group has one or you can get access to one. I hope this helps you and good luck.

Lateral scratch technique, but also I know groups make and can purchase premade functionalized tips that might be of use possibly.
