Surface profilometer will work for your sample.
As far as *surface roughness* is concerned, one needs to scan an *area* or a *length* over the sample surface. This means, the size of sample is important. For sand particles, both size and shape are highly random. So, before choosing an instrument, the sample size should be clarified. Next is to decide the *length scale* and/or *resolution* of measurement. Any statistical roughness parameters (including fractals) are not free from length scale effects. Further, if tactile instruments are used (e.g. stylus method or AFM), the size of probing *tip* interferes with the *resolution*. This can be avoided if non-contact (e.g. optical instruments) are used. So, the process of measuring roughness of sand particles can be as follows:
1/ Decide size of the sand particle to be probed.
2/ Decide the length scale/ resolution of measurement.
3/Decide tip size if tactile (stylus/AFM) methods are used.
4/If possible, measure roughness parameters at least at two length scale.

) and it's bibliography may give more idea on length scale effects.
