I want to measure the root volume of ramie plants grown in pots. I have replicated the trial, and I want to calculate the root volume of each plant in each replication. The procedure is unclear, and I want to get the expert's suggestions and guidelines. Any easy way to do this?
You must wash out the roots as careully as possible over a fine sieve to catch any detached roots
Think of this. The volume of the root system is related to the weight of the root system. So think of weight of the root system as a proxy for the volume of the root system.
Use coarse substrate that will detach from the roots themselves.
Use a time sequence of 2 4 8 weeks sampling plants at each time to get a curve.
Use at least 4 to 8 plants for each time. Soak the system gently remove substrate. Weigh the fresh weight of the system and than dry to get the dry weight.
Make sure you have a both a control and treatments according to your hypothesis.
