If you are interested in the activity of a few well defined enhancer regions then wet lab assays are the way to go. However, if you are interested in a broad range of regions then ChIP-Seq would be a better choice between the two approaches.
ChIP-seq is the way to go. Targeting H3K27ac and H3K4me1 will give you location and activity of enhancers. It can be combined with ChIP-seq on CTCF and ATAC-seq for open chromatin map if you can afford.
