I am checking cardiotonic activity & to perform in vitro study I need to measure cAMP level, for that I am searching for simple techniques which can be performed at laboratory level or best Kit which can be utilized to measure same. So can anybody suggest me such technique or Diagnostic kit (with lowest possible prize & its source)?
In the past I have used the LANCE cAMP 384 kit in my Biochem Pharmacol paper of 2009. This worked fine for us.
L929sA cells were induced as indicated for 300, after which cells
were detached from the plate and counted. Per condition, 6000
cells were analyzed in triplicate using the LANCE cAMP 384 kit
(PerkinElmer, Turku, Finland), according to the manufacturer’s
instructions. Quantification of signal was performed on
a VICTOR3 multiplate reader (PerkinElmer, Turku, Finland).

In the past, we used a radioactive method to detect cAMP on extracts obtained from cell homogenates by means of a commercial kit. To prepare the extracts, you must use speed-waac machine. Pay attention to adjust pH of cell homogenates before using speed-vaac, but do not enlarge too much the volume of samples otherwise it will be impossible to extract the nucleotide. if the samples will be prepared correctely, The method will work very well.
