Hello All,

I have 9 different strains of Pseudomonas that all are positive for HCN prodcution. For qualitative assay, I did the simple plate assay with filter paper saturated in sodium picrate solution. All filter papers turned brownish red from yellow, indicating positive test results.

Now, I want to quantify HCN production from these bacteria. For that, Literature has many reports using Lambert et al., 1975 method.

My question is that how can i make potassium cyanide standard curve and in which concentrations? Some papers report 14 microgram/ml HCN prodcution that does not sound true to me. For example, if I make standard curve with concentrations of 4-20 microgram/L, the results with my sample should show the prodcution of HCN in microgram/L also? Please also help in making a simple KCN standard curve.
