I agree with Pierre Beguin. Better to avoid using HgCl2 due to its high toxicity and disposal problems. I'm surprised that a reference published as late as 2018 is recommending this method. The reference Dr. Beguin provided should work well. Another option is to use agar in which casein is incorporated instead of gelation. The agar w/ casein will be opaque. If the microbe growing on casein agar makes protease, you'll see clear zones around the colonies on an opaque background, and without adding any chemicals. Use of casein agar for protease detection is described in any good microbiology lab manual. It's an old procedure, but it works.
