what are the best methods to make fusarium and pythium inoculum? I read in many papers that water suspenssion is "enough" because these two little fella make spores everywhere !! is it ok to make a culture on PDA then make a water suspension before final inoculation ?
Hi Waffa: For Fusarium I prefer to grow it on millet seed and use it as inoculum , put for Pythium the perfect medium to get height sporulation is to grow it in liquid medium prepared from the same host you isolate the fungal from it, example if you isolate the pythium from cucumber the perfect medium is to grow it on cucumber fruit.

I guess so inoculum preparation of Fusarium is kind of same way like Rhizoctonia on barley/Maiz seeds !! I say this because I am not sure if we have millet seeds in the "seeds market" therefore are there other alternatives (like wheat) specially for Fusarium solani !!

For Fusarium, you can prepare the inoculum by put the sterile D.W. on Petri dish that contains a full growth of Fusarium, then you scratch the media and collect the spores.
For Pythium, I agree with Oadi
There are two ways to prepare fusarium inoculum, first one one using corn media (1/3 sand, 1/3 corn, water) and after autoclaving them , we can inoculate them, and finally they add to soil by 5% which will used for plan growing. second one by preparing PDA broth and inoculate them winch fusarium isolate and after a week u have to calculate the percentage u will use for inoculation (about 100000 spore/ml)
