I am starting my PhD studying the interractions between macrophages and the tumoral extracellular matrix and I am trying to design an experiment to assess the effect of the stiffness of the collagen I scaffold on macrophage polarization and on their way of interracting physically with the collagen fibers.
I have collagen I from bovine skin (C4243 Sigma) and I would like to make gels with different stiffness but I have no idea which concentration of collagen to use to make a soft or a stiff collagen gel...
Do ou have any idea or a starting point ? :)
I plan to assess stiffness with AFM later on
