As far as I know, before performing a biodegradation study of a compound, it is necessary to acclimatize the bacterial culture to that compound (especially if the compound is recalcitrant). Then bacterial growth is monitored by measuring cfu periodically. n numbers of subculturing are done until the growth curve or better say the lag phase time period is approximately same for the (n-1)th and the nth subcultures. Are there other criteria to ensure whether a bacterial culture is acclimatized to a compound? Please reply me your views.

You could say it is acclimatized to a culturing condition when you obtain a stable and maximum growth in such media over aprox. three rounds of culturing to fresh media. Not necessarily similar to the maximum growth obtained in optimal media (like accumulation of toxic metabolites, etc). Max. growth in a given condition could also be assessed by following OD600nm curves. To see if it is stable, by taking a known amount of OD cells at same curve point to fresh media and measure again the growth curve.

I share your views about this and I can also tell Prasenjit that apart from working with the CFUs/ml of bacterial cultures, he can also work with the turbidimetric methods and monitor the optical density of his cultures at regular intervals. By this, he will also be able to determine when the culture stabilizes..that is when there is no longer a net increase in the optical density of the cultures.
