I recommend to break down mechanically while isolating the fibroblast-like cells while performing cell isolation from nasal tissue.
I put a small piece of tissue into a cell-culture petri dish and drop a small amount of complete DMEM onto it, then I make small pieces (minced) of tissue with surgical blades, but you need to break it up really well. In this way, after incubating for 2 hours in the cell culture incubator, I cultivate with freshly prepared complete cell media.
I have tried this product previously: https://www.stemcell.com/easysep-mouse-epithelial-cell-enrichment-kit-ii.html
From my experience using mouse lung tissue, although I got the lung epithelial cells, the yield was very low. Hope to share any experience or recommendation.
