To isolate adequate amount rice protoplasts for PEG mediated transformation you need enough starting material (around 10- 15 gram leaves). Your plants must not be more than 7 – 10 days old and grown with proper light and humidity (important ) under growth chamber condition. Attaching a paper that I followed for my experiment related with protoplasts isolation and transformation.
