We are facing problem with inducing sporulation in F graminearum cultures we have tried SNA, 1/2PDB,Oat Meal, Mung beam and CMC media but the cultures failed to sporulate they we forming round ascus ball like structures can anybody give suggestions what is the problem with my culture

Hi Nagab, you can use carnation leafs to induce sporlation on Fusarium spp.
or just use the same substrate that you isolate the fungus from as a media to grow it, that will be factor tho induce sporlation.
Sporulating media are PDA and CZA . Check pH which play imp role in sporulation. Side by side try to mix subtrate from where fungus isolated with PDA or CZA.
if you use the liquid medium as PDB, you have to put the erlens on top of a rotary incubator with chosen speed like for example 200 r.p.m / mn
You could try Bilai's medium modified by Joffe:
1 g potassium dihydrogen phosphate (KH2PO4)
1 g potassium nitrate (KNO3)
0.5 g magnesium sulphate (MgSO4.7H2O)
0.5 g potassium chloride (KCl)
0.2 g starch powder
0.2 g glucose
0.2 g sucrose
15 g agar
1 litre distilled water
Autoclaving for 20 minutes at 120°C.
Strips of pure cellulose lens paper are aded before the agar had set.
