I have read different protocolos both fast and slow instillation methods and using various volumes of bacterial suspension. I would need to know a standard protocol for intranasal instillation with good recovery of bacteria in lung.
I have read a publication from my colleague about quantification of an intranasal instillation in mice. It is important to adhere to the steps reported by his lab here PMID: 11880310. Generally, you can read that the relative distribution to the lung parenchyma increases with dose. Here in this text 50ul is the good installation volume, after which there is not an increase of distribution, that is e.g., using a 75ul. Mouse needs to be in supine position to deliver compound intranasally (drops into both nares using a pipetman (model P20 or P200, Gilson), please see the M&M. It is a very nice report of installation with good results. You might adjust your method(s) but you have to keep in mind that smaller amounts of installations will have lesser presence in the lung parenchyma.
