To induce salinity stress in agricultural fields for plant growth analysis, several key strategies must be followed. Begin by selecting a field with naturally high salinity levels or add salt to the soil to reach the desired salinity level, keeping in mind that different salts may have varying effects on plant growth. Conduct soil tests to establish baseline salinity levels and make adjustments accordingly. Manage irrigation carefully, using saline water to gradually increase soil salinity while monitoring soil conductivity with electrical conductivity (EC) meters. Adjust the frequency and volume of irrigation to control salt accumulation. To further induce salinity stress, apply salts like sodium chloride or calcium chloride in a controlled manner. Ensure proper drainage to prevent waterlogging and salt accumulation at the root zone. Choose plant species or varieties known to be sensitive to salinity for your analysis. Conduct multiple replicate trials to account for variability in soil and environmental conditions. Regularly measure plant growth parameters and physiological responses, such as chlorophyll content and electrolyte leakage, and analyze the data to assess the impact of salinity stress on plant growth and physiology. Maintain a control group of plants under normal soil conditions for comparison. Be prepared to implement salinity mitigation techniques if salinity levels become excessively high during the experiment, and always adhere to ethical and environmental research guidelines.
we usually develop salinity studies in tomato at production stage in a greenhouse (in big containers for each three plants), and we apply the salt concentrations trhough our irrigation system. One line of plants in control conditions and other in different salinity conditions (50, 100 mM of NaCl). We prepare the salinity solution by adding NaCl to the irrigation water.
