Hi everyone, I'm currently doing DNA extraction for samples from the family Hesperiidae. I've done the extraction and my concentrations range between 1.5 to 4.98 ng/uL, which is quite low. I noticed that during lysis, my samples did not lyse completely, meaning there are still tissues left, although I've left them to incubate for more than 16 hours (overnight incubation). I used the Qiagen DNeasy extraction kit. I was wondering if there's any protocol that I should modify to increase my DNA yield or is it because my samples are small, thats why the concentration is low?
