HI, if you want to remove most of the spores you can centrifuge the extract you have obtained (I imagine you will use a mixture of organic solvents to extract the aflatoxins). If you want to be sure that there are no spores in the extract to be analyzed, you could filter it using 0.45 or 0.20 micrometer filters.

Abstract

Within this research project, the LCI (Lebensmittelchemisches Institut des Bundesverbandes der Deutschen Süßwarenindustrie e. V.) conducted systematic studies to determine the thermal stability of the mycotoxins ochratoxin A (OTA) and aflatoxin B1, as the available literature provides contradictory data. Firstly, the said mycotoxins in pure form were subjected to thermal treament. Secondly, tests were conducted to determine the influence of certain matrix substances, including carbohydrates and proteins, on the thermal decomposition behaviour of the mycotoxins. As a result it can be said that OTA seems to be stable up to 180 °C; however aflatoxin B1 was almost completely degraded at heating temperatures of 160 °C and above. In several model assays it could further be shown that the degradation of mycotoxins is improved by the existence of certain matrix compounds.

Aflatoxin extraction procedure will inactivate the spores. If you extract with 80% or 70% methanol with a brief sonication, you are good to go

Hi greetings to all mycotoxin researchers. I have worked mycotoxins and on that experience i believe Dr. Oadi given nice suggestion. Also you may try heat-cold shock treatment.

Not only aflatoxins are heat resistant but these can also remain stable at as low as -20 degree celcius. I practically observed.

You can use autoclave at a temperature of 121 C and pressure of 15 p/inch2 for about 20-30 minutes. In this method all spores of Aspergillus flavus are killed, while aflatoxins are not affected, because of their thermal stability.

The temperature degree of autoclave which was about 121 C don't degrade the thermal stable aflatoxins which need about 180 C or more for degrading.

The opinion suggested by Shubhransu Nayak (heat-cold shock treatment) is also can be applied. But I see that the autoclaving method is more effective than this method (It's only an opinion).

Autoclaving/heat. Any heat effective vs fungal spores will begin to degrade mycotoxin. You might be able to use heat if you don't need quantiative recovery but you'll need to titrate

Solvent extraction may well obtain mycotoxin. Fungal spores are hydrophic and may well "float" on polar solvent and be unaffected. Binary mixtures of hydrophilic and hydrophobic solvents might work if they can extract mycotixin and suspend fungal spores to prevent dispersion.

I see a lot many suggestions to inactivate spores by heating or autoclave prior to aflatoxins quantification. Aflatoxin B1 is quite stable at autoclave temperature but what about aflatoxin Gs (G1 and G2)? They aren't very stable and tend to degrade readily.

Heating the colonies (which on the culture medium in Petri dishes) continuously by autoclave for several minutes kills the mycelium and spores, and does not break down most of the aflatoxins that need high temperatures to break them down.
