When an ShRNA Knock down efficiency is not good, how I should improve this as I know ShRNA KD efficiency is not dose dependent.. it is meaningless to increase the amount of ShRNA right?
I revised some articles discussing the issue and wanna point out the following points:
1. Higher RNAi concentration will cause higher transfection efficiency but not higher inhibition rate.
2. Higher RNAi concentration will result in off-target effects.
3. Of note, the mentioned article considered the inhibitory rate by RT-qPCR only, which may not be the ideal way in this case and can give miss-leading information.
Here, if you would like to achieve higher inhibitory rate, pls consider different intervals and multiple transfection (e.g: 24h, 36h and 48 and repeat the transfection with same intervals).
