I have to immobilize an antibody onto a paper (preferably nitrocellulose) to develop a paper based immunosensor. Can anyone suggest a reliable and reproducible method of doing so without loosing the orientation of the antibody, so that it can capture the target analyte ?

Few antibodies are immobilized directly by physical absorption onto the nitrocellulose. The literature reports suggest this to be due to the specific hydrophobic and dipolar interaction between the domains of protein and nitrocellulose and this, in turn, leads to preservation of orientation. As for other proteins hope the article is of some use to you.

The paper Aditya send is very important.However need to have specific explanation how to modifies the surface of material. See cross-link chemistry. Chitosan for examples is an atractive biomaterial used in nanotechnology. Chemical as EDC or sulfoNHS is so important.
