Hi Raksha, Good to see you in this network. I wish I could tell you some information. But please understand that I do not have much expertise on this area. Firstly, I would like to ask you that what exactly are you obliged to carry out in Arabidopsis. Because it doesn't matter if you are interested in leaf or other parts of this plant, but however, it would always be best to grow seeds firstly in a petri dish containing corresponding growth medium which is solidified with agar or agarose I guess. As you know that seeds after intense sterilization needs a good solid medium for shoot and root growth. This method I have preferred especially for Genomic DNA isolation in transgenic Arabidopsis when I was doing my thesis training. And the method was good for PCR amplification to find trangene inserts. So, I hope this would help you but I am afraid if it is not up to your expected one.

i would say hydroponic system. Works fine at my lab. Just place A thaliana seeds in an abosrbent material with acces to Hydroponic medium and you are done
I work with 7 day old seedlings mostly grown in petri dish with MS media culture. Growing 21 day-old seedling in these petri dishes is an option too. It gives you enough tissue as working material
Hello! In my group we use 2 different approach according to what you need; 1. grow them is petri dish with ms media and 0.8% agar and then transfer them to soil. (when starting to grow the first true leaves). Or then just sow them directly into soil. To avoid fungi and other contamination we sterilize the seeds. We let them stratify in the dark at 4ºC in water at least 48h. If you need the sterilization protocol let me know. After 4-5 weeks you should have a beautiful rosette full of leaves to use.
we don't have a hydroponic system, but it is very nice if you want to control the exact amount of nutrients that your plants have.
