We are having a problem with contamination in our cell culture. Since a couple of months we are getting frequent but isolated contaminations with bacteria (we think it might be E. coli). We cultured the bacteria and we know that they are sensitive to kanamycin but resistant to pen/strep and ampicillin. The weird thing is that sometimes the contamination occurs in just 1 well of a 12-well plate or in just 1 dish of a stack of 10 cm dishes that contain the same cells/media etc. We have cleaned the entire cell culture room, cleaned the hoods and incubators and changed the HEPA filters. We are changing the water in the water bath and the incubators once per week and we spike it with antibiotics/antimycotics. We are all wearing lab coats and cleaning the surfaces with ethanol before and after working in cell culture. The contamination occurs approximately once per week and we don't know what to do. Does anyone have experience with such a contamination and would know how we can get rid of this bug? Any suggestions are appreciated! Thanks!
