I have few live cell images, and I wanted to study the transporting pattern (as in retrograde and anetrograde). Can someone please tell me how to do this with a kymograph. I use ImageJ, is there a special plugin for it? or do i have to use a different program algorithm for it, if so can someone help.
The ImageJ function dynamic reslice (found under the menus Image>Stacks>Dynamic Reslice) can be used once you draw a single line.
Sometimes for trafficking assays, it would be useful to use a kymograph of more than a single pixel, and that can be done with the make montage function. (Image>Stacks>Make Montage...). For this, you duplicate a stack of the area you wish to have a kymograph of and run the montage on that new stack. Set the number of columns to be the size of your stack and the number of rows to be 1. Make sure border width is 0 and the result will be a multipixel wide kymograph.
