Dear Suganaya,

I have always used Poly-D-Lysine to coat my flasks, especially if the flasks will be shaken to remove the microglia and oligodendrocytes. Just make sure to wash the flasks with sterile water two or three times before plating the cells.

Hi Suganya, I also coat my flasks with poly d lys to seed astrocytes. As Jill have said take care to wash it properly with sterile water and let the flask get dry before seed the cells. We use poly d lys at 1 mg/ml (commercial from Sigma)

Dear Suganya,

We also use PDL at 1 mg/ml, and dissolve it in sterile water. I have not found it necessary to dissolve PDL in pH 8.3 Borate buffer for astrocytes.
