Hi!

Electrophoresis 2005, 26, 501–510 reported that biotin-streptavidin can be fully, reversibly disassociated in de-ionised water at elevated temperature @ ~70℃. I tried to repeat with 20um streptavidin magnetic bead and biotin-FAM probe, in 70℃ DEPC water. However around 70% fluorescence instead of 100% is removed for the first time, and the second binding is just 80% of the first time binding. Is there something missing in my experiment setting?
