Hi everybody
When I get fresh frozen tissue (without fixating with PFA or formaldehyde, just snap frozen with OCT) sliced with the cryostat mounted in a slide it doesn't immediately get attached to the microscope slide so, how do you perform a 95% Ethanol fixation of the slices in the slides prior to further downstream protocols like staining or whatever? You just drop a few drops of 95% Ethanol into the section and let it air-dry over the slide?

What type of glass slides do use?
We always use poly-L-lysine coated glass slides from Thermo (polysine slide) though other brands are available too.
The frozen tissue (4 - 14 um) easily attaches. This can be air dried first. If you want to fix it with ethanol: incubate the slides in -20°C EtOH for 10 minutes. Air-dry and rehydrate in PBS.
