I am having an issue with the pump pressure, it is going upper the limit and I am not sure what's going on. I am pretty sure it is not the column, it is the pump. Is there any advice you can give me about what might be the problem and how to fix it? i tried to clean it and set it up but it did not work
First, contact an experienced chromatographer who is familiar with the specific instrument you are using for help. *These systems takes many years of full time use to achieve a basic level of proficiency with and you probably do not have that much free time to dedicate to this one thing. Students are not expected to know how to do these things. Seek local help, but here are some tips. - Troubleshoot the system's flow path to find the source of the clog. Remove the column, install a clean restriction capillary to obtain the required back-pressure on the system. Flush with an appropriate solution that is 100% soluble with all of the compounds you have been recently running through it. If stable, record the pressure for comparison later. If you find the pressure increasing, then start disconnecting parts, one-by-one, starting at the waste side of the detector moving towards the pump until you find the source of the clog. Replace the clogged part, then flush and re-test.

Sometimes an increasing back-pressure is completely normal. We have no idea what you are doing so should not assume anything. We need very basic info to offer useful advice. Make and model of HPLC. What flow rate are you using (mL/min)? What are the exact HPLC column dimensions and column type (ID x Length, Particle size. Name/brand of column). What is the composition of the mobile phase you are using (names and composition, exactly)?
