Dear Respectable Scientists,
I have been trying to extract DNA from above mentioned sources using different CTAB/SDS Buffer Compositions and purifying agents but I am not able to extract DNA from roots of :
- Dactylorhiza incarnata
- Bergenia ciliata
- Smilax china
- Nannoglottis hookeri
And Barks like:
- Cinnamomum verum
- Santalum album
And Galls like:
- Tamarix indica
- Quercus infectoria
- Pistacia chinensis
I want to use DNA EXT Kits except DNEasy that are a little cheap and please recommend any.
Please share your valuable experience to solve my problem.

I cannot help on the best method but your old sample dna may be slightly degraded and small size. While it may not be visible on gels because it is a smear and not a clear band . Pcr is very sensitive and you may have enough dna for pcr or nested pcr while you get the production sorted out. Try pcr on a couple of samples if that is where your project is going next

I am not able to extract any DNA from the plants. The gel is always perfectly fine as I run the ladder too.
And yes I have to perform PCR but first I cant extract DNA from the plants.

Yes Muhammad Saad my point is that old dna is often randomly broken up into short lengths so when you try to see dna on a gel the small amount of dna is spread over the whole length of the gel and not concentrated into one single visible band.It may be there but not visible. It may still be worth trying the pcr because pcr needs very little template dna and amounts not visible on a gel can be plenty for amplifying with pcr

With due respect my target for DNA EXT is plants.
This method is suitable for it. You can modify the Salt Method, which is very easy to use and cheap.
