I already done run experiment Optical density versus time for two types of microbe against dragon fruit peel extraction (as file attached). From this graph, Is it correct for its shape/pattern? what else I can explain through this graph? from this graph I can said my solution extraction have very low antibacterial activity. I said that because, I ever run for inhibition zone test, the result is negative (clear zone is not appear)

I'm not concentrate my sample due to worried it will degrade some of the active compound that can contribute to the antibacterial activity. In the same time I also study mineral content and phenolic content from solution extraction. after that, antibacterial study are studied on the dragon fruit peel extraction. Bacteria are used e.coli and s.aureus. how about my result? is there have any bacterial effect based on the data?

If I'm not mistaken even we using the temperature below at 50 C, but for sure the time to concentrate the sample will be extended, either use high temperature or long time evaporation, the active compounds will degrade too..i just using water as solvent extraction. the solution extraction are studied for mineral content, antioxidant activity and phenolic compound. the same solution extraction is used to study antibacterial activity. Inhibition zone as file attached (no clear zone for e.coli and s.aureus plate). why some of the antibacterial activity plant extract also has not inhibit the bacteria of Staphylococcus? Based on this graph, antimicrobial still have in my sample right? but just low effect

Thanks for your suggestion. the water solvent use is only 50mL, the most of the journal that i read, they concentrate their sample if their use the solvent in large volume. based on this Od vs time graph as file attached, Can i said my sample contain antibacterial activity with low effect? that's why no clear zone on the plate, Do you agree with me? based on question no 3, if I use 50C to evaporate my solution extraction to remove the solvent, it will take 1 day or half day to complete the process, some of the active compounds (mineral contents and phenolic contents) that i wanna study might be degraded, but i never compare before and after apply with rotary evaporator
