I simulated a dipole antenna which is located vertically to the ground (the axis of antenna is z) in CST(Picture number 1). And the axial ration is calculated/plotted by CST as well(Picture number 2).
1)How can calculate the Axial ratio values by hand?
2)Without knowing the E-filed direction(for example for different kinds of antenna), How can understand the antenna is vertically or horizontally polarized? (one person told me by checking the E-filed values it is possible to find out the polarization kind- it is not acceptable to me).
3)How can clarify the antenna is Left/right hand polarized for circular polarization in CST?

Change it to circular polarization in the far-field properties (right click).
Your antenna has infinite axial ratio - it is linearly polarized.
Unless you understand dB V/m at 1 m, plot gain or directivity instead.
Read the help files on far-field or circular or polarization.

1)Yes,In theory the antenna has infinite axial ratio, but in reality non-ideal dipole antenna has a high but finite value. So is not it possible to calculate it by hand?
2)Could you let me know "Change it to circular polarization in the far-field properties (right click)." what is going to do and also "Unless you understand dB V/m at 1 m, plot gain or directivity instead"?

CST has a question mark on the top bar that gets help.
Right click on farfield in the 2D/3D results in the tree, and choose farfield plot properties.
You asked about circular polarization.
For direction of e-field if you set e-field monitors you can display an arrow plot of field direction. Post processing allows you to plot field components.
